What Is KPV?
KPV (Lys-Pro-Val) is a C-terminal tripeptide fragment of alpha-melanocyte stimulating hormone (alpha-MSH). Despite being only three amino acids long, KPV retains the anti-inflammatory properties of the full alpha-MSH molecule while lacking its melanogenic (skin-darkening) effects. This makes it a valuable research tool for studying inflammation without confounding pigmentation changes.
Mechanism of Action
KPV exerts anti-inflammatory effects through multiple pathways. It inhibits NF-kB nuclear translocation, reducing transcription of pro-inflammatory cytokines including TNF-alpha, IL-1beta, and IL-6. The peptide also modulates inflammasome activation and has been shown to reduce oxidative stress markers in inflammatory models.
Unlike many anti-inflammatory agents, KPV appears to work intracellularly, entering cells and directly interfering with inflammatory signaling cascades rather than blocking surface receptors.
Inflammatory Bowel Disease Research
KPV has shown particular promise in IBD research models. Studies in colitis models have demonstrated reduced mucosal inflammation, decreased inflammatory cell infiltration, improved epithelial barrier function, and reduced disease activity scores.
