What Is DSIP?

Delta Sleep-Inducing Peptide (DSIP) is a naturally occurring neuropeptide first isolated from rabbit brain in 1977. It consists of nine amino acids (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) and is found in the hypothalamus, limbic system, and peripheral tissues. Despite its name, DSIP’s biological roles extend well beyond sleep induction.

Sleep Architecture Research

DSIP has been shown to promote delta wave sleep (slow-wave sleep, stages 3-4) without suppressing REM sleep. This is significant because many pharmacological sleep aids suppress REM or alter normal sleep architecture. DSIP appears to normalize sleep patterns rather than simply inducing sedation.

Stress Response Modulation

Research has revealed that DSIP modulates the hypothalamic-pituitary-adrenal (HPA) axis, reducing cortisol and ACTH levels under stress conditions. This stress-buffering effect may partially explain its sleep-promoting properties, as HPA axis hyperactivity is a common cause of insomnia.
